HI...I was wanting to know if I remove my barrel do I have to readjust my sights when I reassemble it ?

*Should* not have to.
There are other factors which may adjust your POI a bit but not much and often not noticeable. Things like barrel gap, barrel indexing, and torque of the barrel nut.
Of course, switching loads/ ammo brands can have a greater affect. So, break it down and clean away- BA removal is our advantage...
Technically, the glass is always full; half liquid, half air....
